پيوند سلول هاي بنيادي مزانشيمي بند ناف باعث كاهش پيشرفت FSGS در موش هاي صحرايي مي گردد

هدف: مطالعه اثر محافظتي سلول هاي بنيادي مزانشيمي بند ناف انسان (HuMSCs) بر پيشرفت FSGS (focal segmental glomerulosclerosis) در موش هاي صحرايي.

 روش ها: HuMSCها به روش كشت چسبنده، كشت داده شدند. موش هاي صحرايي Sprague Dawley به صورت تصادفي در سه گروه طبقه بندي شدند. هيچ عملي بر روي موش هاي گروه كنترل نرمال صورت نگرفت. گروه مدل و گروه پيوند تحت تزريق درون وريدي ماده آدريامايسين با غلظت 4 mg/kg در روز اول و غلظت 3.5 mg/kg در روز هشتم قرار گرفتند.  علاوه براين، به موش هاي گروه پيوند يك ميلي ليتر از سوسپانسيون HuMSCها (2*10(6):/ml) در روزهاي 1، 8، 15 و 22 از طريق سياهرگ دم تزريق شد درحاليكه موش هاي گروه مدل به همان ميزان و با روشي مشابه، DMEM دريافت كردند.

 نتايج: پيوند HuMSCها به طور قابل توجهي ترشح پروتئين اوره و هيپوآلبومينميا را کاهش داد، سطح سيستاتين C را کم نمود و آسيب كليه و نفوذپذيري سلول هاي التهابي در بافت كليوي را بهبود بخشيد. علاوه براين، HuMSCها بيان TGF-β1 و mRNA فاكتور رشد بافت پيوندي (CTGF) در كليه و سطح IL-6 و TNF-α سرم را كاهش دادند.

نتيجه گيري: پيوند HuMSCها توانست پيشرفت FSGS را در موش ها تعديل يا مهار كند. مكانسيم عمل آن ها ممكن است به تنظيم سنتز و آزادسازي حدواسط هاي التهابي و مهار نفوذ سلول هاي التهابي مربوط باشد.

Abstract

Objective To study the protective effect of human umbilical cord mesenchymal stem cells (HuMSCs) on the progression of focal segmental glomerulosclerosis (FSGS) in rats. Methods HuMSCs were incubated by adherent culture. Sprague Dawley rats were randomly assigned to 3 groups: No intervention was applied to the rats of normal control group. Model group and transplant group were injected with adriamycin 4 mg/kg (on day 1) and 3.5 mg/kg (on day 8) via the tail vein, in addition, the transplant group rats were injected with 1 mL HuMSCs suspension (2×10(6);/mL) via the tail vein on day 1, 8, 15 and 22, whereas the model group rats were injected with the same dosage of DMEM in the same way. Results Transplantation of HuMSCs remarkably alleviated urine protein excretion and hypoalbuminemia, decreased serum cystatin C level and improved kidney damage and inflammatory cell infiltration in renal interstitium. Furthermore, HuMSCs decreased the expressions of TGF-β1 and connective tissue growth factor (CTGF) mRNA in kidney and reduced the levels of serum IL-6 and TNF-α. Conclusion Transplantation of HuMSCs could alleviate or inhibit the progression of FSGS in rats, of which the mechanism might be related to the regulation the synthesis and release of inflammatory mediators and the inhibition of inflammatory cell infiltration.
