Due to their important roles in development and cancer, the mechanisms of HS-regulated Shh release and spreading to responding cells are of great interest. We recently found that Shh is released from the producing cells by ADAM (A Disintegrin And Metalloprotease) mediated shedding. Based on this release mechanism, we plan to investigate the following:
- How is Shh release from producing cells regulated?
- Shh is released in clustered form. Are these clusters homo-or heteromultimers? What is their structure? How is their size regulated?
- How do these clusters spread through the extracellular matrix?
The applicant will hold a Masters degree and has gained first biochemical research experience. Experience in Drosophila experimentation is highly welcome, but not required. The successful applicant will find strong support within the excellent interdisciplinary environment of the first Indo-German Research and Training group in Molecular and Cellular Glyco-Sciences (MCGS).
methods:
A wide range of biochemical methods will be employed, such as recombinant mutant protein production and chromatographic/functional characterization, determination of Shh/HS binding (Biacore), and in vivo testing of any obtained models of protein release and clustering.
