ماتریکس خارج سلولی اثرات بیولوژیکی ملاتونین را در سلول های بنیادی مزانشیمی تنظیم می کند

هر دو نوع ویژگی خود تجدیدی و تمایز ویژه دودمانی سلول های بنیادی مزانشیمی در داخل بدن توسط ریزمحیط آنها  از جمله ماتریکس خارج سلولی (ECM) و هورمون های مترشحه به راه می افتد. ماتریکس خارج سلولی ممکن است عملکرد فیزیولوژیکی هورمون ها را با ارائه جایگاه های اتصال و تنظیم مسیرهای سیگنالینگ پایین دست  تنظیم کند. بنابراین، هدف از این مطالعه بررسی میزان جذب ملاتونین رسوب ماتریکس خارج سلولی سلول طبیعی و اثرات این تعامل درعملکرد های بیولوژیکی ملاتونین در سلول های بنیادی مزانشیمی مشتق از مغز استخوان انسان (BM-MSC) بود. ریزساختارهای فیبریلی، ترکیب ماتریکس، و میل ترکیبی ملاتونین ماتریکس خارج سلولی فاقد سلول شده تعیین ویژگی شد. رسوب ECM  سلول ،  تکثیر سلولی بواسطه ملاتونین را تا 31.4٪ بهبود ، تجمع گونه های اکسیژن فعال داخل سلولی را کاهش و بیان mRNA و پروتئین آنزیم سوپراکسید دیسموتاز  (SOD) را افزایش داد. تعامل با ECM به طور قابل توجهی اثرات استخوان زایی ملاتونین را در سلول های بنیادی مزانشیمی مشتق از مغز استخوان با افزایش رسوب کلسیم تا 30.5٪، تنظیم بیان ژن ویژه استئوبلاستی و کاهش بیان متالوپروتئاز (MMP)بهبود داد. مکانیسم های زیربنایی این تغییرات بیان ممکن است با دخالت آنزیم های آنتی اکسیدان داخل سلولی باشد، چرا که در حضور مهار کننده های ویژه SOD  ژن های ویژه استئوبلاستی کاهش بیان یافته بود، در حالی بیان MMP افزایش یافته بود. در مجموع، یافته های ما اهمیت ECM طبیعی را در تنظیم اثرات القای استخوان زایی و آنتی اکسیدانی ملاتونین نشان داده و یک الگوی جدید را برای مطالعه عملکرد بیولوژیکی عوامل رشد و هورمون ها در ریز محیط فیزیولوژیکی مربوطه ارائه می کند. علاوه براین، درک بهتر از افزایش رشد و تمایز استخوانی سلول های بنیادی مزانشیمی توسط ترکیبی از ECM و ملاتونین می تواند طراحی یک جایگزین پیوند برای مهندسی بافت اسکلتی را بهبود بخشد.

Extracellular matrix modulates the biological effects of melatonin in mesenchymal stem cells.

Abstract

Both self-renewal and lineage-specific differentiation of mesenchymal stem cells are triggered by their in vivo microenvironment including the extracellular matrix (ECM) and secreted hormones. The ECM may modulate the physiological functions of hormones by providing binding sites and by regulating downstream signaling pathways. Thus, the purpose of this investigation was to evaluate the degree of adsorption of melatonin to natural cell-deposited ECM and the effects of this interaction on the biological functions of melatonin in human bone marrow-derived mesenchymal stem cells(BM-MSCs). The fibrillar microstructure, matrix composition, and melatonin-binding affinity of decellularized ECM were characterized. Cell-deposited ECM improved melatonin-mediated cell proliferation by 31.4%, attenuated intracellular reactive oxygen species accumulation, and increased superoxide dismutase (SOD) mRNA and protein expression. Interaction with ECM significantly enhanced the osteogenic effects of melatonin on BM-MSCs by increasing calcium deposition by 30.5%, up-regulating osteoblast-specific gene expression, and down-regulating matrix metalloproteinase (MMP) expression. The underlying mechanisms of these changes in expression may involve intracellular antioxidant enzymes, because osteoblast-specific genes were down-regulated, whereas MMP expression was up-regulated, in the presence of SOD-specific inhibitors. Collectively, our findings demonstrate the importance of native ECM in modulating the osteoinductive and antioxidant effects of melatonin and provide a novel platform to study the biological actions of growth factors or hormones in a physiologically relevant microenvironment. Moreover, a better understanding of the enhancement of MSC growth and osteogenic differentiation by the combination of ECM and melatonin can improve the design of graft substitutes for skeletal tissue engineering.
