PhD Position in Identification of Leptin Transporters at the Blood-Brain Barrier (Lubeck, Germany)

The DFG-funded Graduiertenkolleg 1957 studies the effects of hormones derived from adipose tissue (adipokines) on CNS function and in turn, the control of adipose tissue and body weight by the brain. We are looking for highly motivated candidates to join the following PhD project from 01.05.2017 (or earlier).

Project Description:

The adipokine leptin acts on the brain to limit food ingestion. In obesity this feedback mechanism is impaired, probably because the transport of leptin over the blood-brain barrier is reduced. However, the molecular mechanisms of leptin transport over the blood-brain barrier are largely unclear. We will study leptin transport in an in-vitro model of the blood-brain barrier. To identify the genes that are involved in the transport, we will perform siRNA screens of candidate transporters. Perspectively, we will modulate the expression of leptin transporters in vivo and evaluate the effects on body weight and metabolism.

In vitro labeling of peptides
Transwell assays of peptide transport
Generation of AAV vectors
Handling and phenotyping of mice
Western blotting, immunohistochemistry, PCR, qPCR

Qualifications
- Research-based MSc degree passed with an above-average grade in natural sciences, preferably in Biology, Molecular Life Science, Neuroscience, Nutrition & Biomedicine, Pharmacology or a related subject
- Excellent English language skills in speaking and writing
- Pro-active attitude, good communication skills and be able to work effectively in an interdisciplinary team
- Previous research experience in the proposed topic is of great advantage
