PhD position "The role of human Cdc14 phosphatases"

The group of E. Schiebel (http://www.zmbh.uni-heidelberg.de/schiebel/default.shtml) is looking for a highly motivated PhD student with a strong background in biochemistry, cell biology or molecular biology. The successful candidate will be part of an international, highly motivated team of PhD students and postdocs that works at the forefront of scientific research. The PhD student will be a member of the Hartmut Hoffmann-Berling International Graduate School of Molecular and Cellular Biology (HBIGS; http://www.hbigs.uni-heidelberg.de/). The position (E13/60%) is funded for 3 years with the possibility of extension. The applicant will be a member of the SFB1036.

Method::
Cdc14 belongs to a family of highly conserved dual-specificity phosphatases that are present in a wide range of organisms from yeast to human cells. Human Cdc14A has a role at centrosomes. Cdc14B is implicated in DNA damage checkpoint activation, DNA repair and aging 1-4. However, little is known about the substrates of hCdc14A and hCdc14B and their regulation. The goal of this thesis is to study the regulation of hCdc14A and hCdc14B, to identify substrates and to unravel phosphatases that may function together with hCdc14A and hCdc14B. The successful candidate will perform siRNA screens using human Cdc14A/B knockout cell lines for phosphatases with functions that overlap with hCdc14A/B. Cdc14 entrapment mutants will be used to identify substrates. The role of the substrates will be studied by a broad method spectrum including construction of gene knockouts in mammalian cells (ZFN and CRISPR/Cas9), live cell imaging, cell cycle analysis and the analysis of phospho-mutants.
