دیابت حاملگی عملکرد سلول های بند ناف فرزندان در جهت سلامت قلبی عروقی بهبود می بخشد

بیماری های غیر واگیردار مانند دیابت شیرین نوع دو ریشه در تکوین جنینی و مشکلاتی مانند دیابت بارداری مادر(GDM) دارند، ما قصد داریم این فرضیه را در سلول های اولیه مشتق از  نوزادان مادران GDM در مقایسه با کنترل تست کنیم. ما سلول های اولیه مشتق از بند ناف مانند سلول های اندوتلیال سیاهرگی بند ناف انسانی(HUVECs) و سلول های بنیادی مزانشیمی مشتق از ژله وارتون(WJMSCs) را هم در فرزندان مادران GDM و  هم مادران غیر GDM ارزیابی کردیم. ما در ارزیابی های مبتنی بر سلول ما تکثیر، مصرف اکسیژن میتوکندریابی و توانایی حمایت رشد عروق خونی را ارزیابی کردیم. ما مطالعات میکرواری بیان ژنی به همراه آنالیز مسیرهای بعدی و اعتبار بخشی ژن کاندیدا را انجام دادیم. ما تفاوت های قابل توجهی مانند نرخ متابولیک پایین و اختلال در تشکیل لوله اندوتلیالی را در سلول های با پیش زمینه GDMمشاهده کردیم. سلول های HUVEC مشتق از افراد با مادر مبتلا به GDM، بیان پایین تری از پروتئین های ضد آپوپتوزی BCL-xL داشتند که حاکی از به خطر افتادن توان رگزایی است. آنالیزهای بیان ژنی مقایسه ای، تشکیل عروق خونی را به عنوان مسیر اصلی تقویت شده در HUVECهای مشتق شده از GDM با مارکر سطحی CD44 به عنوان ژن بارز بیان شده در گروه GDM نشان داد. اعتبار بخشی عملکردی CD44 نشان داد که آن ها تشکیل لوله ها را در HUVECها تنظیم می کنند و به موجب آن دیدگاه های جدیدی را در مورد مسیرهای جدید ایمپرینت شده در سلول های مشتق از بند ناف اولیه مشتق از فرزندان GDM ارائه می دهند. داده های ما نشان می دهد که سلول های اولیه جداسازی شده از بند ناف فرزندان مادران مبتلا به GDM، ایمپرینت های متابولیکی و مولکولی هایپرگلیسمی مادری را حفظ می کنند که خطر افزایش یافته بیماری های قلبی عروقی را در مراحل بعدی زندگی منعکس می کنند.

Gestational Diabetes alters functions in offspring's umbilical cord cells with implications for cardiovascular health.

Abstract

As non-communicable diseases such as type 2 diabetes mellitus have their roots in prenatal development and conditions such as maternal gestational diabetes (GDM), we aimed to test this hypothesis in primary cells derived from the offspring of GDM mothers compared to control subjects. We have assessed primary umbilical cord derived cells such as human umbilical vein endothelial cells (HUVECs) and Wharton's jelly derived mesenchymal stem cells (WJMSCs) from both, the offspring of GDM and non-GDM mothers. We have compared the primary isolates in cell based assays measuring proliferation, mitochondrial oxygen consumption, and the ability to support blood vessel growth. We conducted gene expression microarray studies with subsequent pathway analysis and candidate gene validation. We observed striking differences between the two groups such as lower metabolic rates and impairment of endothelial tube formation in cells with GDM background. HUVECs from subjects with maternal GDM have lower expression of the anti-apoptotic protein BCL-xL suggesting compromised angiogenic capabilities. Comparative gene expression analysis revealed blood vessel formation as a major pathway enriched in the GDM-derived HUVECs with the surface marker CD44 as a significant gene under-expressed in the GDM group. Functional validation of CD44 revealed that it regulates tube formation in HUVECs thereby providing new insights into a novel pathway imprinted in primary umbilical cord-derived cells from GDM offspring. Our data demonstrate that primary cells isolated from the umbilical cord of offspring born to GDM mothers maintain metabolic and molecular imprints of maternal hyperglycemia reflecting an enhanced risk for cardiovascular disease later in life.
